I find it hard to take this all too seriously. I buy a moderately expensive supplement, and then i hear no good unless you buy more, buy more and buy more, all of which are nice earners for you. So my “follow the money” logic cuts in and makes me suspicious. Is this just a marketing scam?

According to Chris Masterjohn taking a too high dose of Nicotinamide can deplete Methyl groups, because they are spent getting rid of the excess. And he recommends TMG to pervent problems from that.
That the salvage path can produce too much NAM, more than the cells can handle, is new to me.
That should be less of a problem, I think. I guess theres no research showing this directly.
Many manages high doses of B3 without TMG. But the risk is there. Some regard TMG as an insurence.
The CD38 story is very complicated. You don’t want to get rid of it. But it is a big consumer of NAD+.
So, Apigenin could be beneficial, sometimes. It probably is. But getting rid of the scenescent cells producing it would be a good thing. Better, although difficult.
A little Apigenin from Parsley, perhaps dried, instead of a supplement might be a compromise.
So, the story isn’t a scam. The mechanisms are there. But to get it right might not be so easy.
One way to handle it is not to take those very large doses of B3.
And use regular exercise to keep the NAD+ system healthy.
Exercise probably the most important ingredient for that anyway.
Just look at the peer-reviewed journals buddy. Then you don’t have to project your opinions that aren’t vetted in your own research

APIGENIN & QUERCETIN is the combination I’m going with instead of NMN.
Short Story, Back in the 80’s my mother had breast cancer the size if a fist. For some reason she started craving parsley (Apigenin), she never craved parley before, or after. She would have my dad buy crazy amounts, she would sit in bed each night, and eat a large green Tupperware bowl worth of parsley for months. This was before Apigenin was a thing, I guess she just knew what her body needed. Long story short, the cancer shrunk into a dead cell, the doctor removed it, Complete remission, and her experience ended up in some medical books apparently.

These patents involve a variety of uses including the preparation of antiviral

From my humble readings on the topic, these are indeed some POTENTIAL downsides still subject to further investigation through further clinical trials – NOBODY knows yet whether there is truth in this. Therefore a more appropriate title in my opinion would be “NMN & NR could potentially be harmful unless…”, rather than “NMN & NR WILL harm you unless…”. David Sinclair’s says that he takes TMG “out of an abundance of caution”. Mr Black has apparently been spending time investigating, consulting and writing about anti-aging for many years, but I find no qualifications or accepted scientific publications from him. So I would treat this video as an interesting opinion of a well-read person, but not more (and many YouTubers dramatise a bit for extra clicks). In other words, don’t worry unduly if you have been taking NMN without the above additional supplements – and by the way, there is literature indicating that taking Quercetin daily without breaks may also be detrimental.

According to him it’s wise to take TMG if you take large doses of any form of vitamin B3.
About half the dose of that of NMN.
The reason for that is that the body consumes metyl groups when getting rid of the excess.
This is simple biochemistryand could easily be verified.
I’m pretty certain that 1 g doses of NMN is regarded as an excess by the body. That is, if it’s bioavailable enough to get into body.
There’s some controversity about that.
Well, all the negatives coming out about NMN incl the 12 or so human studies are puzzling if indeed we believe Dr Sinclair.. And the Metro Biotech NMN, which caused the FDA ban, has not materialized yet but time will tell.

Great presentation and adding new angle to understanding to this topic. After few years of following Dr.Sinclair I came to conclusion that we should be way, way more cautious with conclusions and experimenting. People without deep vested interest in biotech could be much safer source than ambitious scientists that discovered the power of money.
Now I take Fisetin which is very similar to Quercetin / is Fisetin an acceptable alternative for the NAMPT problem ?
DoNotAge need to put together a pill with a combination of NMN, TMG and smaller amounts of Berberin, Quercetin, etc. Basically a combo of all the latest/current thinking wrt. Lifespan enhancement. Although somewhat experimental and with an exact formula that may change, Iām sure it would be a winner.
More money in separation…. LOL! But that would be great!!!
